Fifty people is a crowd, and crowds need simple rules. Anything with more than two or three instructions will not survive being passed across that many people, and the version played at the far end of the hall will not be the version you described.
Demonstrate rather than explain. Ten seconds of showing what the game looks like beats a minute of describing it, and it works regardless of whether people at the back could hear you.
Expect to need helpers — one per team is ideal, and they need briefing before the group arrives, not during. Expect equipment to go missing. Expect the whole thing to take longer than it did with twenty.
What works at this size: games where everyone moves at once, games with teams large enough that individual performance does not decide the outcome, and games where being out lasts seconds rather than minutes.
